Effects of fatty acids on lysis of Streptococcus faecalis.

Palmitic, stearic, oleic, and linoleic acids at concentrations of 200 nmol/ml all inhibited autolysin activity 80% or more in whole cells or cell-free extracts. This concentration of the saturated fatty acids palmitic acid and stearic acid had little or no effect on the growth of whole cells or protoplasts. However, the unsaturated fatty acids oleic acid and linoleic acid induced lysis in both situations. This lytic effect is apparently not related to any uncoupling activity or inhibition of energy catabolism by unsaturated fatty acids. It is concluded that unsaturated fatty acids induce cell and protoplast lysis by acting as more potent membrane destabilizers than saturated fatty acids.

A synthetic medium has been developed in which Streptococcus faecalis var. zymogenes X-14 elaborates a lysin. The medium consists of 18 amino acids, lactose, six vitamins, adenine, guanine, uracil, and six salts. Glucose, and K(2)HPO(4) in excess of 0.5% (wt/vol), were inhibitory to lysin production; increasing concentrations of l-arginine-hydrochloride gave increasing yields of lytic activity.

Addition of the antibiotic cerulenin to cultures lowered the minimal effective concentration of penicillin G or methicillin required to produce bacterial lysis and killing. This effect was most pronounced at subinhibitory antibiotic concentrations. Cerulenin had no significant effects on lysis or killing induced in the presence of D-cycloserine, fosfomycin, bacitracin, or vancomycin.
